Molecular Cloning and Cell Culture techniques learned to apply in research of looking Bax gene to relate its’ function with apoptosis in cancerous cells with following details:
• Performed RNA isolation, followed by RT-PCR, and Gel validation. (Invitrogen Kit).
• Designed primers for “Bax gene”, followed by RT-PCR and PCR to amplify the selected gene using another Invitrogen kit; PCR purification by spin-column technology implementing QIA quick PCR purification kit.
• Enzymatic digestion using Eco-RI and KpnI to ligate Bax gene into PC DNA 3.1 vector, followed by Gel Validation.
• Heat shock transformation of plasmid construct into DH5α cells and conducted Mini Prep assay after bacterial cultures; verification of colonies for transformation with EcoRI and KpnI digestion and Gel validation.
• Accountable for aseptic techniques of Mammalian cell culture of PC 3 cell lines, sub-culturing, splitting, harvesting, and freezing; transfection of PC 3 cells with Bax gene and actin control with EX Gen 500 reagent.
• Western blotting analysis for protein expression purified by SDS-PAGE, protein assay for concentration using Bio-rad kit.
• Visualization of immuno fluoresced HeLa cells using immunostaining technique.
• Familiar with flow cytometry.
Genetics Lab
• Cell division studies; calculation of seed exercise segregation ratio and goodness of fit for mono- and dihybrid crosses; chromosomal mutation analysis using UV rays; Barr bodies demonstration; induction of polyploidy; Chi-square analysis; biostatistics tests; immunoassays; DNA-RNA-protein purification and estimation; isolation of auxotroph by replica plating method; PCR; tissue culture; electrophoresis; etc.
