A. CLEANING, RINSING, EQUILIBRATING, STORING AND SHUTTING DOWN THE LC-100
  1. Cleaning the LC-100:

    1. If the system has been stored in 0.1 M NaOH, then proceed to part 2. If not, place all four buffer lines into 0.1 M NaOH.
    2. Ensure the outlet tubing from the injector is connected to the inlet of the detector. If not connect appropriate tubing in place.
    3. Turn on the instrument. Press "MENU".
    4. Select "FLUSH", enter.
    5. The LC-100 will flush the entire system (buffer lines, proportional valves, pumps and system tubing) with 0.1 M NaOH. When the system stops, repeat steps c-d. Once flow stops, leave the system sitting in caustic for 10 minutes.

  2. Rinsing the LC-100:

    1. Place all four buffer lines in sterile filtered deionized water and the waste line in a beaker.
    2. Turn on the instrument if not already on and press "MENU".
    3. Select "FLUSH", enter.
    4. Repeat steps b and c until the water exiting through the waste line tested with pH paper is £ 7.0. This ensures that all caustic has been removed from the system. The system is ready to be equilibrated.

  3. Equilibrating the LC-100 system for use:
    1. Place all four buffer lines in Equilibration buffer ( Tris-HCl, pH 6.0).
    2. Turn on the instrument. Ensure the outlet tubing from the detector is attached to the inlet tubing of the detector.
    3. Select "MENU".
    4. Select "FLUSH", enter.
    5. Repeat steps 2 &3.
    6. System is ready. See section C for protocol on Equilibrating the column

  • Storing and shutting down the LC-100:
    1. Place all four buffer lines in 0.1 M NaOH. Ensure the outlet of the injector is hooked up to the inlet tubing of the detector.
    2. Press "MENU"
    3. Select "SHUTDOWN". This will flush caustic through the system at 10 ml/min for four minutes. Make sure you have enough caustic supply since no air should enter the system.
    4. After system stops, follow prompt for correct way of shutting down the instrument.
    5. Place a status tag on the instrument with the Name: LC-100

          Status: Stored
          USE by Date: one month from today
          Step: 0.1 M NaOH
          Date/ Initials:_______________

    6. Turn off the instrument and discard waste liquid appropriately.
