- Make sure panel is closed and cuvette holders are empty before starting.
- Turn spectrophotometer "ON" by flipping the yellow switch on the side.
- The machine will automatically initialize and make a baseline correction.
- Select "8" CONDITION SET, then press ENTER.
- Select "5" for LAMP SELECT to turn off UV light bulb, then press ENTER.
- Press the FILE key.
- Select "3" for CELLS (E.coli), then press ENTER.
      "7" for PICHIA P (yeast)
      "8" for CHO cells (animal)
- It will then ask you for "Parameter Change Y/N", Select NO, and press ENTER.
- Fill 2 cuvettes each with 2.2 ml of blank solution. When holding the cuvette, please hold it from the line up or it will tamper the measurements. Wipe cuvettes with Kim wipes.
- Open panel door.
- Place blank cuvettes in cuvette holders.
- Press AUTO ZERO key.
- Remove blank cuvette from front holder and relace with sample to be tested. Close panel door.
- Press START to take a reading.
- Remove sample from front holder. Leave machine on for future readings. Only turn off when you are certain that you are done with it.
- To test additional samples: Place cuvette in front holder and press START for reading. Do not replace cuvette blank in rear holder.
- Press COPY for a hard copy/printout.
- Press RETURN to bring you back to Step 8.* (This will erase your old data.)
- Press FILE to return to the original screen.
- Remove cuvettes remaining in holders.
- Flip power switch located on the side to turn the machine off.
