  1. Make sure panel is closed and cuvette holders are empty before starting.

  2. Turn spectrophotometer "ON" by flipping the yellow switch on the side.

  3. The machine will automatically initialize and make a baseline correction.

  4. Select "8" CONDITION SET, then press ENTER.

  5. Select "5" for LAMP SELECT to turn off UV light bulb, then press ENTER.

  6. Press the FILE key.

  7. Select "3" for CELLS (E.coli), then press ENTER.
          "7" for PICHIA P (yeast)
          "8" for CHO cells (animal)

  8. It will then ask you for "Parameter Change Y/N", Select NO, and press ENTER.

  9. Fill 2 cuvettes each with 2.2 ml of blank solution. When holding the cuvette, please hold it from the line up or it will tamper the measurements. Wipe cuvettes with Kim wipes.

  10. Open panel door.

  11. Place blank cuvettes in cuvette holders.

  12. Press AUTO ZERO key.

  13. Remove blank cuvette from front holder and relace with sample to be tested. Close panel door.

  14. Press START to take a reading.

  15. Remove sample from front holder. Leave machine on for future readings. Only turn off when you are certain that you are done with it.

  16. To test additional samples: Place cuvette in front holder and press START for reading. Do not replace cuvette blank in rear holder.

  17. Press COPY for a hard copy/printout. 

  18. Press RETURN to bring you back to Step 8.* (This will erase your old data.)

  19. Press FILE to return to the original screen.

  20. Remove cuvettes remaining in holders.

  21. Flip power switch located on the side to turn the machine off.
